Active Ribosome profiling
Active ribosome profiling or RiboLace, is based on a proprietary puromycin-analogue molecule capable of isolating only actively translating ribosomes through a fast and simple antibody and tag-free pull-down approach. This method offers a more reliable way to assess the translational state of any biological system, showing higher concordance with protein levels compared to classical ribosome profiling.
Ribosome profiling
Ribosome profiling involves sequencing mRNA fragments protected by ribosomes which are isolated using canonical techniques like sucrose cushion or polysome profiling. This technique offers a comprehensive view of translation with the unique ability to provide information about ribosomes positioning along transcripts at single-nucleotide resolution within living cells.
Polysome profiling
Polysome profiling relies on sucrose density gradient centrifugation to separate translating mRNAs (ribosome-bound) from those that aren't being translated. The following sequencing of ribosome-bound mRNA transcripts (Polysome-Seq) enables determination of translation activity.
